"""""主要从事肿瘤代谢与DNA损害修复的研究。主要学术成就如下:\r\r1、PI3K通路对肿瘤糖酵解的调节。发现非依赖于AKT的、PI3K调控的肿瘤糖酵解在肿瘤细胞内起着重要作用,打破了传统认为PI3K调控肿瘤糖酵解主要依赖于AKT的认识。以前的研究认为AKT是PI3K主要的下游分子,但AKT在肿瘤中突变率总体较低,同时临床上使用AKT抑制剂治疗肿瘤的效果明显不如PI3K抑制剂,原因不清楚。我的研究表明PI3K通过Rac--醛縮酶通路调节糖代谢可能更为重要。我们发现位于糖代谢通路中下游的醛縮酶是整个肿瘤糖代谢的限速酶,而AKT的调节点主要是糖代谢上游的糖类磷酸化酶。我们的研究还发现这一调节方式实际是通过调节细胞骨架重构实现的,这表明细胞骨架重排和糖代谢是偶联在一起的。我们知道,几乎所有的细胞行为都涉及到细胞骨架重排。因此,细胞通过将细胞骨架重排和糖代谢偶联,就能及时保证细胞行为有充分的能量和生物质供应,这是一种最简单又最有效的调节方式。这是对糖代谢特别是肿瘤糖代谢调节方式认识的重大突破。临床上泛PI3K抑制剂对肿瘤治疗效果明显,但由于其明显抑制正常细胞葡萄糖吸收,大多数病人不能耐受。我的研究表明,PI3K-RAC-醛縮酶这一调节通路主要通过PI3K-α异构体起作用,在肿瘤中PI3K-α是PI3K突变的主要异构体,由于细胞中通常还有其他PI3K的异构体,单独抑制PI3K-α并不明显影响正常细胞葡萄糖吸收,但对活跃的肿瘤糖代谢起到明显的抑制作用。根据这一研究结果,我们提出了用PI3K-α抑制剂替代原来药物组合中的泛PI3K抑制剂的治疗策略,用以治疗三阴性乳腺癌。一期临床的初步研究中表明,其效果明显优于原来的组合,并且副作用明显下降,这使得PI3K抑制剂在临床应用中前进了一大步。我们研究还阐明了为何肿瘤中PI3K-α是PI3K突变的主要异构体,也解释了AKT抑制剂临床治疗效果差强人意的原因。该课题主要由Lewis Cantley,Gerburg Wulf教授指导,相关研究的文章以被Cell和PNAS杂志接收发表。\r\r2、EB病毒在乳腺癌发生中的作用。确认了EB病毒感染乳腺上皮细胞的途径。发现EB病毒感染乳腺上皮能够促进乳腺干细胞扩增,从而促进乳腺癌的发生。该课题由直接导师Gerburg Wulf教授指导。在完成EB病毒研究工作的同时,帮助Gerburg Wulf教授成功申请到AVON基金肿瘤病毒研究的课题。相关研究的文章以被eBiomedicine杂志接收\r\r3、Pin1蛋白在肿瘤细胞DNA损伤修复中的作用。发现Pin1通过调节Brca1参与DNA同源重组修复。确认了Pin1与Brca1的相互作用方式及作用位点。 "近期论文
