1. 重要天然药物及其生物合成基因(簇)资源的发掘 结合比较基因组学、化学生物学等多学科手段,充分发掘具有自主知识产权与中国特色的微生物及植物等来源的重要天然药物及其生物合成基因(簇)资源,积极参与国际竞争。 2. 重要天然药物合成途径的解析与改造 综合分子遗传学、生物化学、有机合成化学及结构生物学手段,对活性显著、结构独特的重要天然药物的生物合成机理进行探究,以破解自然界中神奇的化学与生物学之谜;进而利用合成化学与代谢工程手段对目标天然药物进行修饰改造,以创造出活性更高,副作用更低的理想目标药物。 3. 重要药物(中间体)的合成生物学制造及其产业化开发 以国民经济与市场需求为导向,选择重要天然药物(中间体)为研究对象,从头设计合成模块,并进行组装调试,以重构成最优化合成途径,实现目标天然药物(中间体)的高效合成生物学制造,进而实现其产业化开发。与此同时,与著名生物制药企业紧密合作,以工业菌株为研究材料,深入解析目标药物合成途径与代谢网络,通过合成生物学策略,理性化提高目标药物(有效组分)的产量和质量,从而直接服务于工业化生产。近期论文
